Z offset set to -50 still not printing

I have set my z offset to -50 and I am still unable to print.

Do I have to adjust my speed? Temperature?

What am I doing wrong? Am I missing something?

I just watched an instructional video and set my Z offset to -145. The PLA stuck along the side test however when it began printing the object it was still gathering the PLA loosely and the shape was too large as well it was rounded instead of being rectangular with 90 degree corners.
